Adenine and Guanine Nucleotide Metabolism During Platelet Storage at 22OC

چکیده
Adenine and guanine nucleotide metabolism of platelet concentrates (PCs) was studied during storage for transfusion at 22 f 2°C over a 7-day period using high-pressure liquid chromatography. There was a steady decrease in platelet adenosine triphosphate (ATP) and adenosine diphosphate (ADP), which was balanced quantitatively by an increase in plasma hypoxanthine. As expected, ammonia accumulated along with hypoxanthine but at a far greater rate. A fall in platelet guanosine triphosphate (GTP) and guanosine diphosphate (GDP) paralleled the fall in ATP + ADP. When adenine was present in the primary anticoagulant, it was carried over into the PC and metabolized. ATP, GTP, total adenine nucleotides, and total guanine nucleotides declined more slowly in the presence of adenine than in its absence. With adenine, the increase in hypoxanthine concentration was more rapid and quantitatively balanced the decrease in adenine and platelet ATP + ADP. Plasma xanthine rose during storage but at a rate that exceeded the decline in GTP + GDP. When platelet ATP + ADP was labeled with 14C-adenine at the initia-

The effect of platelet concentrate storage temperature on adenine nucleotide metabolism.
The effect of platelet concentrate storage temperature (4 degrees C versus 22 degrees C) on platelet adenine nucleotide metabolism was studied. In general, levels of platelet ATP and ADP, the release reaction, and the metabolis nucleotide pool were best preserved for 72 hr by storage of concentrates at 4 degrees C. Storage of concentrates for 72 hr at 22 degrees C was occasionally associated wi...

We have evaluated the NMR shielding tensors for A:T,G:C,A:U in Watson-crick, and U-quartet. We have computed NMR shielding tensors at B31YP level by using 6-31G(d) basis set. We have compute anisotropy and asymmetry in A:T,G:C,A:U and U-quartet. The NMR shielding tensors were calculated using the GIAO method. The natural bonding orbital analysis (NBO) were performed. NBO calculation have been ...
متن کاملThe Effect of Benzyl Adenine on Qualitative Characteristics and Antioxidant Capacity of Plum Fruit during Storage
In order to evaluate the effect of Benzyl Adenine (BA) on qualitative characteristics and shelf life of plum fruits CV “Gold Drops”, an experiment was conducted as a factorial based on randomized complete block design with three replications. In this experiment different concentrations of BA (0, 50, 75, 100 and 125 mg L-1) were applied as a foliar application. After harvesting, fruits were stor...

Background: The purpose of the present in vitro study was to evaluate the effect of long term storage of conventional platelet concentrates (PCs) on major platelet receptors CD42b and CD41/CD61 by flow cytometry method and also measuring the overall platelet procoagulant activity status using platelet factor 3 (PF3) assay. Materials and Methods: Six random units of conventional platelet conce...

At a concentration of 9.6 x 10(-5)M, 2,6-diaminopurine (DAP) completely inhibited cell enlargement, cell division, and DNA synthesis (determined by microphotometric measurement of Feulgen dye) in Vicia faba roots. Inhibition of cell enlargement was partially reversed by adenine, guanine, xanthine, adenosine, and desoxyadenosine. Guanine and the nucleosides gave the greatest reversal, suggesting...
